In Vitro Selection of Salt - Resistant Solanum tuberosum L . Varieties

Abstract
In vitro salt resistance screening of 20 potato varieties was carried out using culture medium with 5 different levels of sodium chloride. Responses of these varieties to different salt concentrations were determined by studying rooting of stem segments, increase in fresh weight, length of roots as well as shoots and microtuberization. Normal shoot growth was observed in the medium containing low concentration of NaCl (0.05 M), on the other hand, the medium containing high concentration of NaCl (0.5 M) showed no growth of shoot. Differences in growth of potato varieties were observed in the range of 0.1M and 0.2M salt medium among the various treatment concentrations used in this study for in vitro selection of salt-resistance varieties. As a result of in vitro screening, 10 varieties such as Atlantic, Bintje, Desiree, Dejima, Kennebec, La Rouge, Norchip, Red La Soda, Red Pontiac, and Russet Burbank were selected as relatively salt resistant varieties from the present experiment. Continuous field experiment at salty soil condition is necessary for agricultural application of salt resistant potato varieties selected from in vitro screening system.
